چکیده

A vector derived from an original infectious clone of Turnip mosaic potyvirus (TuMV) has been developed. The vector (p35Tunos-vec01) was made through the creation of a new unique insertion site for foreign genes between the NIb and the CP genes of the viral genome. The jellyfish Green Fluorescent Protein (GFP) and E. coli β-glucuronidase (GUS) were expressed in Arabidopsis thaliana plants infected with chimeric vectors carrying their corresponding genes. In the case of GUS, expression levels that were 15-50 fold higher than nuclear transgenic A. thaliana lines carrying the same gene were attained. Acceptable levels of gene stability were found in the infected plants, compatible with the use of the vector for protein production in plants. A non-aphid transmissible version of the vector was also made and its lack of aphid transmission was extensively shown. The vector was not transmitted through seeds or by contact between plants. Additional key words: aphid transmission, Arabidopsis thaliana, biofactories, environment, potyvirus.

Transient expression of green fluorescent protein in radish (Raphanus sativus) using a turnip mosaic virus based vector

It is possible to use transgenic plants, as bioreactors, for the production of recombinant inexpensive chemicals and drug components. Transient gene expression is an appropriate alternative to stable transformation because it allows an inexpensive and rapid method for expression of recombinant proteins in plant tissues. In recent years, plant viral vectors have been increasingly developed as su...

Replication and encapsidation of recombinant Turnip yellow mosaic virus RNA.

Turnip yellow mosaic virus (TYMV) is a positive strand RNA virus that infects mainly Cruciferae plants. In this study, the TYMV genome was modified by inserting an extra subgenomic RNA promoter and a multiple cloning site. This modified TYMV was introduced into Nicotiana benthamiana using a Agrobacterium-mediated T-DNA transfer system (agroinfiltration). When a gene encoding beta-glucuronidase ...

Utility of P19 Gene-Silencing Suppressor for High Level Expression of Recombinant Human Therapeutic Proteins in Plant Cells

Background: The potential of plants, as a safe and eukaryotic system, is considered in the production of recombinant therapeutic human protein today; but the expression level of heterologous proteins is limited by the post-transcriptional gene silencing (PTGS) response in this new technology. The use of viral suppressors of gene silencing can prevent PTGS and improve transient expression level ...

Targeting of the turnip yellow mosaic virus 66K replication protein to the chloroplast envelope is mediated by the 140K protein.

Turnip yellow mosaic virus (TYMV), a positive-strand RNA virus in the alphavirus-like superfamily, encodes two replication proteins, 140K and 66K, both being required for its RNA genome replication. The 140K protein contains domains indicative of methyltransferase, proteinase, and NTPase/helicase, and the 66K protein encompasses the RNA-dependent RNA polymerase domain. During viral infection, t...

Expression and Purification of Human Interferon Gamma Using a Plant Viral Vector

A plant viral vector engineered from an in vivo infectious clone of zucchini yellow mosaic virus(ZYMV) was used to express the human interferon-gamma (INF-γ) in planta. The INF-γ gene was in frame inserted between the P1 and HC-Pro ORFs of the ZYMV vector. The infectious activity of the vector was approved by rubbing the plasmid on Chenopodium quino a and observing local lesions. Individual les...
